CIA induction kits

For induction of collagen-induced arthritis (CIA) in DBA/1 mice

Supplied in pre-filled syringes, ready to use.

CIA is induced by chicken or bovine collagen antigen in CFA, followed 18–21 days later by collagen in IFA.

  • Consistently induces arthritis in 90–100% of DBA/1 mice (Taconic DBA1BO)

When used with the recommended protocol, up to 20% of mice develop arthritis within 16 to 20 days, and the remainder within 30 to 35 days of immunization. Most mice reach disease severityof 10 to 14 (on the scale 0 to 16).

Each lot is tested and individually adjusted to ensure consistent disease induction.

Collagen-induced arthritis (CIA) is one of the most commonly used animalmodels of human rheumatoid arthritis (RA).

Joint inflammation, which develops in animals with CIA, strongly resemblesinflammation observed in patients with RA. Blocking tumor necrosis factor(TNF) isefficacious in treatment of CIA, just as it is a highly efficacioustherapy in treatment of RA patients.

CIA is mediated by both T-cells and antibodies (B-cells). Macrophages arebelieved to play an important role in mediating tissue damage during diseasedevelopment.

CIA is induced by immunization with collagen/CFA emulsion, followed 18–21 days later by collagen/IFA emulsion. See recommended protocol and product details.

While both bovine type II and chicken type II collagen are used for CIA induction, bovine collagen appears to be slightly better at inducing CIA in male DBA/1 mice, while chicken collagen works slightly better in females.

* Breeder substrain can substantially affect CIA development. Hooke recommends use of Taconic DBA1BO mice with these kits.

Each kit provides sufficient emulsion for 20 mice.

Antigen is chicken type II collagen (EK-0210, EK-0211) or bovine type II collagen (EK-0220, EK-0221).

Data are from three independent experiments using Hooke Kits™ Chicken Collagen/CFA Emulsion (EK-0210) and Chicken Collagen/IFA Emulsion (EK-0211), with female DBA/1 mice (Taconic Biosciences).

Similar results are obtained using Bovine Collagen/CFA Emulsion (EK-0220) and Bovine Collagen/IFA Emulsion (EK-0221) in male DBA/mice (Taconic Biosciences).

Frequently asked questions

Is the source of DBA/1 mice important?

CIA development is sensitive to DBA/1 mouse substrain. Hooke recommends use of Taconic Biosciences DBA1BO mice with these kits. Our testing shows arthritis will be consistently induced in 90 to 100% of DBA/1 mice from Taconic. Results may differ in mice from other breeders.

Is the booster dose (collagen with IFA) really necessary for CIA induction?

Yes, in our experience it is necessary. However, mouse substrains can differ, and a very few labs have reported that their mice develop CIA after a single immunization, without a booster. Most labs need a booster dose for good CIAdevelopment. With the booster, disease onset is 5-10 days after the booster in almost all mice. Without the booster, CIA usually develops in only 20-75% of mice, and onset is much more spread over time.

Are there gender differences in CIA development?

We recommend using male mice. CIA develops very similarly in males and females, but females are more likely to develop skin lesions near the injection site; in mice with lesions CIA develops poorly. Males rarely develop skin lesions.

Can these kits be used to induce CIA in Lewis rats?

If Hooke"s protocol is followed, our bovine collagen CIA kits will induce CIA in Lewis rats (both males and females).

Use the same protocol recommended for mice, except at double the dose, so 0.1 mL/rat of Bovine Collagen/CFA Emulsion (EK-0220) for the first injection, and then 7 days later, Bovine Collagen/IFA Emulsion (EK-0221) again at 0.1 mL/rat. At this dose, each kit is sufficient for 10 rats.
