Theundifferentiatedstateofembryonicstem(ES)andinducedpluripotentstem(iPS)cellscanbecharacterizedbyahighlevelofalkalinephosphatase(AP)expressionwhich,alongwiththeexpressionofothersurfaceMarkers,indicatesundifferentiatedcellswiththepotentialtoself-renew.APisahydrolaseenzymeresponsIBLefordephosphorylatingmoleculessuchasnucleotides,proteins,andalkaloidsunderalkalineconditions.WhenfixedESoriPScellsarestainedusingtheAPStainingKit,undifferentiatedcellsappearredorpurple,whereasdifferentiatedcellsappearcolorless.
