Interleukin15isacytokinewithstructuralsimilaritytoIL-2.LikeIL-2,IL-15bindstoandsignalsthroughtheIL-2/IL-15betachain(CD122)andthecommongammachain(gamma-C,CD132).IL-15issecretedbymononuclearphagocytes(andsomeothercells)followinginfectionbyvirus(es).Thiscytokineinducescellproliferationofnaturalkillercells;cellsoftheinnateimmunesystemwhoseprincipalroleistokillvirallyinfectedcells.Interleukin15(IL-15)regulatesTandnaturalkiller(NK)cellactivationandproliferation.SurvivalsignalsthatmaintainmemoryTcellsintheabsenceofantigenareprovidedbyIL-15.ThiscytokineisalsoimplicatedinNKcelldevelopment.Inrodentlymphocytes,IL-15preventsapoptosisbyinducinganapoptosisinhibitor,BCL2L1/BCL-x(L).IL-15hasbeenshowntoenhancetheanti-tumorimmunityofCD8+Tcellsinpre-clinicalmodels.AphaseIclinicaltrialtoevaluatethesafety,dosing,andanti-tumorefficacyofIL-15inpatientswithmetastaticmelanomaandrenalcellcarcinoma(kidneycancer)hasbeguntoenrollpatientsattheNationalInstitutesofHealth.
